A Single Nucleotide Polymorphism in the MDM2 Gene Disrupts the Oscillation of p53 and MDM2 Levels in Cells

P53 and MDM2 protein levels have been shown to oscillate upon genotoxic stress. A mathematical model of P53/MDM2 dynamics predicts that elevated levels of MDM2 would make the oscillation disappear. Independently, Arnie and his group have shown that elevated MDM2 levels are created by SNP309 (T->G), that reinforces a Sp1 binding site. In this paper, the authors show that SNP309 indeed suppresses oscillations. Moreover SNP309 makes MDM2 transcription estrogen-dependent on.
